I thought the application of the Nobel aspiring student was hard to beat. Until just now, I had no idea that application spam existed. For the current PhD student vacancy in our lab for a project concerning the neurogenetics of behavior in octopamine-deficient flies, I just received the CV of a molecular biologist accompanied by this letter without either address, signature or explanation (formatting original):

Well said by Faraday if you will spend all of your time in writing things,  then when you will get time to think about it. I believe that the basic principles and laws of science are always same everywhere in the universe. The centripetal force just balanced by centrifugal, the balance of all gases in atmosphere and the dynamics of genome and arrangement of nucleotides in the genome. Every thing is deliberately just balanced and the interaction among all of these things allows them to work in the right direction(or in any resultant direction) and control their work flow.



My experience says nothing is difficult enough to stop us not to doing that, untill and unless we will not go through it, Nature is unpredictable only when we have no information or data or knowledge about it. As soon as we understand its events we can precisely say that why it  is going on?, where on earth wind doesn’t blow many of the countries are planning to start their wind farm and UK is the first who has launched one of its wind farm to produce 2.5% of its annual electricity.



The poorest country of this world spend its 70% of the total income on education, combodia no longer has its army and like to invest for the maintenance of forest, may be because they are circumspect and thinking about their decendents or childern. Using ground water for the agriculture is not a solution at all, I have seen many people in africa those use the unvaluable drops of water to irregate their crops and living their from thousands of years.



Every idea is tough and seems difficult to execute but greatest engineers out there those are ready to face challenges and accept them with a different smile on their face. I have seen big snakes on the sea shore of france to generate electricity by tidal energy, I have seen  japan is using geothermal energy to generate electricity, denmark no long allowing smoke to enter into the atmosphere which is coming out from its thermal power plant is this the solution for ever or the soluiton for future, no body knows.



Education and our natural resources are the things we have earned in last several thousand  years, how we can let our real treasure destroy  in front of our eyes and are we ready to answer all of the questions , our descendents will ask.  Now a days many of the countries are spending more and more on their education system to make it more strong and to allow their childern to explore the nature and to learn its basic principle which is difficult to find or observe in side the artificial laboratory, instead of this we need a huge work station, which is everywhere arround you “ The Nature”.



DNA sequencing is useful to determine the pattern of nucleotides present in any genomic sequence; there are various ways to determine the nucleotide sequences. One of the methods is Chain termination developed by Frederick sanger, but now a days we are using highly specialized and controlled sequencing techniques such as 454 sequencing. The main problem which has been solved by this research, how could we surmount the problem of repetitive regions. DNA sequencing has solved many problems associated with the genome of microorganisms or macro organisms those are pathogenic or lethal to the human beings and causes disease to them, for example disease like Typanosomiasis caused by Trypanosoma brucei 927, this disease is lethal and cause sleeping sick ness in humans and in few of the domestic animals. The host of this organism is Tsetse fly which injects it into the body of animal and causes disease.



Sequencing of genome is probability based concept, the accuracy of this process depends on the combination of nucleotides present in the 454 artificial reads generated, we have used bimodal graph to shred genome and we have developed one script for that.



Sequencing is very tricky thing to do and I am working on it from last one year this time is enough for me to come up with a recipe to improve the sequencing process and to surmount the problem of repetitive region, I was working on the genome of Trypanosoma brucei927, this genome contain different types of repetitive units such as VSG and ESAG. The most problematic thing about genome is its repetitive elements. Now we know that how we can overcome this problem and how we can assemble the genome in one go.



I believe that the coverage and combination of insert size can gives us a 100% perfect result and in this way we can assemble any of the genome in one go including its sub-telomere region and other highly repetitive region.



We have got 99.99% or 100% alignment for many of the contigs to the reference genome particularly sub-telomere region this the case when we are using combination of insert sizes, for example 6 and 8Kb, for some coverage.



The shredding of the genome is the most important step randomly generated reads can not gives us any idea about the  main problem, therefore the solution is to shred genome by the help of bimodal graph.
